A PCR test for mumps detects the virus’s genetic material (RNA) in samples such as buccal/throat swabs, saliva, or urine. The laboratory amplifies viral RNA to confirm acute infection, especially early in illness around gland swelling. Results usually come within 24–72 hours; a negative test may require repeat testing or paired serology if the sample timing was suboptimal.

Interpretation: A positive mumps IgM or a positive PCR from saliva/throat indicates recent or active infection. A rising IgG titer between acute and convalescent samples shows recent infection; a single positive IgG usually indicates past infection or immunity.
