Human sperm assay

To test for human sperm, provide a fresh semen sample to a clinic or lab for microscopic semen analysis, which measures sperm count, motility, morphology and volume. Home sperm-count kits detect semen concentration but don’t assess morphology. In forensic or clinical settings, swabs can be analyzed for semen markers (acid phosphatase, PSA) and examined microscopically; DNA testing confirms male origin.

The HSA (human serum albumin) test is done by drawing a blood sample from a vein, usually in the arm, by a phlebotomist; fasting is typically not required. The blood is sent to a lab where albumin concentration is measured and reported (g/dL). For kidney checks, a urine sample (spot or 24‑hour) may be collected to measure urinary albumin or the albumin‑to‑creatinine ratio.

Most clinics ask men to produce a semen sample by masturbation into a sterile container; partners may be present for support but should not assist physically or use lubricants. Some clinics accept intercourse-collected samples only with a clinic‑provided non‑spermicidal collection condom. Transport the sample to the lab within one hour at body temperature and follow clinic instructions, including 2–7 days’ abstinence beforehand.

1.5 ml of semen is a small volume — about 0.3 teaspoon (roughly one‑third of a teaspoon), ~30 drops, or about 1.5 grams by weight. Clinically, normal ejaculate volume is roughly 1.5–5 ml, so 1.5 ml sits at the lower end of the typical range and is the usual lower reference value used in semen analysis.

Yes — home sperm test kits can check basic measures (concentration and some motility) and give a quick indication of sperm presence and count. They’re useful for screening but aren’t a full fertility workup: they don’t reliably assess morphology, DNA integrity, hormones, or infections. Abnormal or concerning results, or persistent fertility problems, should prompt formal laboratory semen analysis and clinical evaluation.

Sperm count and overall semen quality are highest in early adulthood, generally peaking in the 20s to early 30s. After about age 40, sperm concentration, motility, and morphology gradually decline, and DNA damage rates increase. Individual factors such as health, lifestyle, smoking, obesity, and some medications also affect sperm count at any age.
