ANA Blot Test

The ANA blot tests for specific anti‑nuclear autoantibodies directed against extractable nuclear antigens (e.g., Ro/SSA, La/SSB, Sm, RNP, Scl‑70, Jo‑1). It confirms positive ANA screening and identifies antibodies linked to autoimmune connective‑tissue diseases such as systemic lupus erythematosus, Sjögren’s syndrome, systemic sclerosis, mixed connective tissue disease and inflammatory myopathies. Results help guide diagnosis and management.

An ANA blot (autoantibody immunoblot/ENA panel) typically costs about $50–$400 (US), £40–£200 (UK private labs), or ₹1,000–₹6,000 (India), depending on test type, lab, and whether it’s a single ANA screen or extended blot/panel. Public systems often cover it when clinically indicated. Ask your lab or insurer for exact pricing.

A positive ANA blot alone doesn’t require treatment; it indicates possible autoimmune disease. Management depends on symptoms and specific autoantibodies: confirm with further testing and clinical correlation, refer to a rheumatologist, and monitor organ involvement. Symptomatic care may include NSAIDs, antimalarials (hydroxychloroquine) and short-course corticosteroids. Moderate–severe or organ-threatening disease may need DMARDs or biologic immunosuppression tailored to the diagnosis.

A positive ANA blot isn't always cause for alarm. It can suggest autoimmune disease (for example, lupus or Sjögren’s), but low titres or isolated antibodies often appear in healthy people, infections, or drug reactions. Meaning depends on symptoms, titre, pattern and specific antibodies. Discuss results with your doctor or a rheumatologist for clinical correlation and further testing; seek prompt care for fevers, rashes, severe joint pain or organ symptoms.

ANA blot (line immunoassay) accurately identifies specific anti‑nuclear antibodies and has high specificity for those targets, but sensitivity is lower than indirect immunofluorescence (IFA). It can miss antibodies not included in the panel, so false negatives occur; false positives are possible but less common. Results must be interpreted with clinical context and, when needed, confirmed by IFA or additional testing.

A positive ANA can occur with certain cancers, most often hematologic malignancies (lymphomas, leukemias) and some solid tumors such as lung, breast, ovarian and hepatocellular cancers. Tumor-related immune dysregulation or paraneoplastic autoimmune phenomena may cause ANA positivity. ANA alone isn’t diagnostic for malignancy; further evaluation depends on symptoms, clinical exam, imaging and specialist assessment.
