ANA Blot Profile - Immuno Blot

The ANA profile immunoblot is a blood test that detects antinuclear antibodies and identifies specific autoantibodies (for example SSA/Ro, SSB/La, Sm, RNP, Scl‑70, Jo‑1) against nuclear and cellular antigens using immunoblot technology. It helps diagnose and differentiate autoimmune connective-tissue diseases (like lupus, Sjögren’s, scleroderma, myositis), guides further evaluation, and should be interpreted with clinical findings and other tests.

A positive ANA blot indicates antinuclear antibodies are present, suggesting immune activity against cell nuclei. It raises suspicion for autoimmune conditions (for example, lupus, Sjögren’s, scleroderma, or mixed connective tissue disease) but is not diagnostic by itself. False positives can occur with infections, medications, or aging. Clinical correlation, specific antibody tests, and specialist evaluation are usually required.

ANA is a screening test (usually indirect immunofluorescence on HEp‑2 cells) that detects and reports the presence, titer and pattern of anti‑nuclear antibodies, useful for identifying possible autoimmune disease. An ANA blot (or ENA/autoantibody panel) is a follow‑up, more specific immunoblot/line assay that identifies antibodies to defined nuclear antigens (Sm, RNP, SSA/SSB, Scl‑70, Jo‑1), improving diagnostic specificity.

Antinuclear antibodies (ANA) can be positive in various cancers, particularly hematologic malignancies such as lymphoma, leukemia and multiple myeloma, and in solid tumors including breast, lung, ovarian, colorectal, gastric, hepatic and pancreatic cancers, as well as melanoma. ANA positivity varies by tumor type, may reflect paraneoplastic autoimmunity or treatment effects, and is nonspecific—not diagnostic of cancer.

A positive ANA alone usually isn’t urgent. Many healthy people—especially older adults—have low-titer ANAs without autoimmune disease. Worry increases with high titers, specific staining patterns, symptoms (joint pain, rashes, fatigue) or abnormal blood tests. Discuss results with your doctor or a rheumatologist for targeted antibody testing and follow-up to determine cause and need for treatment.

The ImmunoBlot test detects specific antibodies or proteins in a patient’s blood, helping confirm or rule out infections and autoimmune conditions. It’s commonly used as a confirmatory assay after a positive screening test—for example, to verify Lyme disease, HIV, hepatitis, or to identify disease-specific autoantibodies—providing more specific identification of antigen–antibody reactions than many initial screening methods.
