ZnT8 antibody is an autoantibody directed against the zinc transporter 8 protein on pancreatic beta cells. Its presence indicates autoimmune beta‑cell damage and is used, alongside GAD and IA‑2 antibodies, to help diagnose and predict type 1 diabetes risk and progression. Testing helps distinguish type 1 from type 2 diabetes and assess autoimmune activity, especially in newly diagnosed patients and at‑risk relatives.

Type 1 diabetes is indicated by autoantibodies against pancreatic beta cells: GAD65 (glutamic acid decarboxylase), insulin autoantibodies (IAA), islet cell antibodies (ICA), insulinoma-associated‑2 antibodies (IA‑2), and zinc transporter 8 antibodies (ZnT8). Their presence confirms autoimmune beta‑cell destruction, helps distinguish type 1 from type 2 diabetes, and can predict future disease risk, especially when multiple antibodies are present.

Zinc transporter 8 (ZnT8, SLC30A8) is primarily linked to diabetes. Autoantibodies to ZnT8 are diagnostic markers of type 1 diabetes and latent autoimmune diabetes in adults. Genetic variants in SLC30A8 influence beta‑cell function and insulin secretion, altering risk of type 2 diabetes (some loss‑of‑function variants are protective). Associations with gestational diabetes have also been reported.

A common laboratory reference for ZnT8 (zinc transporter 8) autoantibodies is that values below about 15 U/mL are considered negative/within the normal range. Some assays report an index rather than U/mL and cutoffs can differ by lab. Results above the lab’s reference suggest autoimmune beta‑cell involvement; discuss any abnormal result with your clinician for interpretation and next steps.

ZnT8 (zinc transporter 8), encoded by SLC30A8, concentrates zinc into pancreatic beta-cell insulin secretory granules. Zinc is essential for insulin crystallization, stable storage, proinsulin processing and regulated insulin secretion. ZnT8 activity therefore supports proper insulin maturation and release; immune targeting or genetic variants can impair its function and influence diabetes risk.

Type 1 diabetes cannot currently be prevented. It’s an autoimmune disease in which the immune system destroys insulin-producing pancreatic beta cells, influenced by genetic and environmental factors. No lifestyle change or medication has been proven to stop its development, though research is exploring immunotherapies and vaccines to delay or prevent onset in high-risk people. Early screening in relatives enables prompt diagnosis and treatment.
