Pap stain (Papanicolaou stain) is used to evaluate cells collected in cytology specimens—most commonly cervical (Pap) smears—to screen for precancerous and cancerous changes. It highlights cellular and nuclear morphology, allowing detection of dysplasia, malignancy, infections, and inflammation. It’s also applied to other body fluids and fine‑needle aspirates to assess cell types and detect abnormal or infectious processes.

Papanicolaou (Pap) stain uses hematoxylin (Harris or Gill) to stain nuclei, plus polychrome cytoplasmic counterstains: Orange G (OG‑6) and the EA mixture (eosin Y and light green SF, sometimes with Bismarck brown). Orange G highlights keratin, while the eosin/green components differentiate cytoplasmic maturation and cell types, aiding cytological interpretation. It is widely used in cervical cytology.
