Giemsa stain (a Romanowsky-type stain) is the standard for malaria diagnosis on thick and thin peripheral blood films; thick films increase sensitivity while thin films allow species identification and parasite counting. Alternative Romanowsky stains such as Wright or Field may be used for rapid or resource‑limited settings, but Giemsa remains the preferred method for routine detection and morphology.

Gram stain classifies bacteria as Gram‑positive (retain crystal violet–iodine, appear purple) or Gram‑negative (decolorize and take a counterstain), based on cell‑wall peptidoglycan; it’s used for bacterial identification. Giemsa is a Romanowsky stain for blood smears, parasites, and intracellular organisms, giving blue‑purple nuclei and pink cytoplasm—used for cell morphology and parasite detection rather than Gram reaction.

Giemsa stain is used on peripheral blood smears and bone marrow aspirates for hematology, on thin and thick blood films to detect malaria and other blood parasites, on cytology specimens (cerebrospinal, pleural, peritoneal fluids, BAL), on touch or impression smears from tissues or lesions, and on some histology/tissue sections and chromosomes for karyotyping.

Giemsa staining visualizes blood and tissue cells to identify blood-borne parasites (notably Plasmodium species causing malaria, Trypanosoma, Leishmania), intracellular organisms and cellular inclusions. It improves assessment of red and white blood cell morphology for diagnosing anemia and leukocyte disorders, aids peripheral smear and bone marrow evaluation, and is also used for chromosome banding in cytogenetic studies.

Giemsa stain is used to visualize blood cells, parasites and microorganisms in clinical and research labs. It highlights red and white blood cell morphology on peripheral blood and bone marrow smears, helping diagnose malaria, babesiosis, trypanosomiasis and other protozoal infections. It is also used for detecting certain bacteria and for chromosome (G-banding) staining in cytogenetics.

The classic stain for tuberculosis (TB) sputum microscopy is the Ziehl–Neelsen acid‑fast stain, which uses carbol fuchsin to stain Mycobacterium tuberculosis that resists decolorization by acid‑alcohol; a counterstain (methylene blue or malachite green) highlights background cells. Fluorescent stains (auramine‑rhodamine) are also used for more sensitive screening. Kinyoun is a cold acid‑fast variant.
