Alpha-2 macroglobulins are large plasma glycoproteins mainly synthesized by the liver that function as broad-spectrum protease inhibitors. They trap and inactivate proteases, modulate inflammation, and bind cytokines and growth factors, helping regulate immune responses and tissue remodeling. Levels change in conditions such as inflammation, pregnancy, nephrotic syndrome (often elevated) and severe liver disease (can be reduced).

Alpha‑2 globulins are a plasma protein fraction that includes alpha‑2 macroglobulin, haptoglobin, ceruloplasmin and alpha‑2 antiplasmin. They act as acute‑phase reactants and perform protease inhibition, bind and clear free hemoglobin, transport copper, and regulate fibrinolysis. Levels change with inflammation, infection, liver disease and nephrotic syndrome, aiding host defense, coagulation balance and metal transport.

Alpha-2-macroglobulin (A2M) is primarily synthesized by the liver (hepatocytes) and secreted into plasma. Extrahepatic cells including macrophages, fibroblasts, and astrocytes can also produce it locally. It functions as a broad-spectrum protease inhibitor and acute-phase protein found in blood and interstitial fluids, with levels changing in inflammation, liver disease, and certain metabolic conditions.

Alpha-2‑macroglobulin (A2M) is primarily produced by the liver and secreted into the bloodstream. Other cells — including macrophages, fibroblasts and certain brain glial cells — can also synthesize it. A2M circulates as a broad-spectrum protease inhibitor and carrier for cytokines and growth factors; its blood levels change with inflammation, acute-phase responses, and some liver disorders.

High alpha-2 globulin means increased levels of certain blood proteins (like haptoglobin, ceruloplasmin, alpha‑2‑macroglobulin) and often reflects acute or chronic inflammation, infection, liver disease, nephrotic syndrome, pregnancy, or some cancers. Alpha-2 (α2) adrenergic receptors are Gi‑coupled receptors that inhibit norepinephrine release, reducing sympathetic outflow. Activation lowers heart rate and blood pressure, produces sedation and analgesia, and can decrease insulin secretion. Centrally it promotes calmness and vasodilation; peripherally it may cause transient vasoconstriction. These effects explain use of drugs like clonidine and dexmedetomidine.
