Qualitative tests for acetone include the nitroprusside (Rothera/Legal) test and the iodoform test. Nitroprusside in alkaline medium yields a purple or magenta color indicating ketone bodies (acetoacetate/acetone). The iodoform test (iodine plus NaOH) produces a characteristic yellow crystalline precipitate (iodoform) with methyl ketones such as acetone. These are screening tests; confirmatory assays measure exact levels.

A positive acetone (ketone) test means your body is producing ketones—byproducts of fat breakdown—because it lacks sufficient glucose for energy. Causes include fasting, low‑carb diets, prolonged exercise, vomiting, illness, pregnancy or insulin deficiency in diabetes.
