Smear for Gonococci, in Pimpri Chinchwad

Testing for gonococci (Neisseria gonorrhoeae) uses nucleic acid amplification tests (NAATs) on urine or swabs (endocervical, vaginal, urethral, rectal, pharyngeal) — the preferred, most sensitive method. Culture from swabs is used when antibiotic susceptibility is needed. In symptomatic men, Gram stain of urethral discharge can show intracellular gram‑negative diplococci. Samples should be taken from exposed sites for accurate diagnosis.

The preferred diagnostic test for gonorrhea is nucleic acid amplification testing (NAAT) of appropriate specimens because of its high sensitivity and specificity. Bacterial culture from urethral, cervical, pharyngeal, or rectal sites is the traditional confirmatory test and remains essential for antibiotic susceptibility testing and outbreak investigation; culture is recommended when treatment failure or resistance is suspected.

Yes. In symptomatic men, gonorrhoea can be diagnosed by finding gonococci (intracellular Gram‑negative diplococci) on a urethral smear by microscopy — a rapid, highly specific test. However, sensitivity is lower in asymptomatic men; nucleic acid amplification testing (NAAT) or culture is recommended for confirmation, antimicrobial susceptibility testing, and when microscopy is negative.

Neisseria gonorrhoeae are Gram‑negative, non‑motile, oxidase‑positive diplococci with a characteristic "kidney‑bean" or "coffee‑bean" shape. They commonly appear in pairs and are frequently seen intracellularly within polymorphonuclear neutrophils on Gram‑stained smears of purulent exudate, though extracellular organisms may also be present. Cultures on selective media form small, convex, grayish colonies; intracellular location aids rapid diagnosis.

Gonorrhea is usually confirmed by a nucleic acid amplification test (NAAT), which detects Neisseria gonorrhoeae DNA or RNA from urine or swabs of the urethra, cervix, throat, or rectum. Culture can also confirm infection and is used for antibiotic susceptibility testing and outbreak investigation. NAAT is the preferred initial diagnostic method because of its high sensitivity and specificity.

A Pap smear screens for abnormal cervical cells and HPV-related changes but does not reliably detect gonorrhea. Gonorrhea is diagnosed with specific tests—nucleic acid amplification tests (NAATs), cultures, or PCR—performed on cervical, vaginal, urethral swabs, or urine. If you have symptoms or exposure risk, ask your clinician for STI testing specifically for gonorrhea.
