What is a ENA Test?
The ENA test looks for antibodies that target specific proteins inside cell nuclei. These antibodies are called extractable nuclear antigen antibodies. They can show that the immune system is mistakenly attacking the body's own tissues. Detecting these antibodies helps find and classify autoimmune connective tissue diseases. Common conditions include lupus, Sjögren’s syndrome, scleroderma, mixed connective tissue disease, and inflammatory muscle disease. Doctors use ENA results with symptoms, physical exam, and other tests to confirm a diagnosis. Results can guide treatment choices and help monitor disease activity over time. A positive result does not always mean disease and needs clinical correlation.

Why Take a ENA Test?
ENA is ordered as part of autoimmune serology panels or after a positive ANA to identify specific antibodies. Doctors may request it when patients have unexplained joint pain, rashes, dry eyes or mouth, muscle weakness, or systemic symptoms. It helps diagnose or distinguish lupus, Sjögren’s, scleroderma, mixed connective tissue disease, and myositis. Abnormal results most often come from autoimmune disease but can be influenced by infections or certain medications, and a family history of autoimmune conditions can make testing more likely.
