CD4 (Pan T Cell Marker)

A pan‑T cell marker is a surface protein expressed on nearly all T lymphocytes (for example, CD3) used to identify and quantify T‑cell populations. It confirms T‑cell lineage in blood and tissue samples, aids immunophenotyping by flow cytometry or immunohistochemistry, and helps distinguish T cells from B cells or NK cells in diagnostic and research settings.

CD4 T cells are CD3+ CD4+ lymphocytes. Subset and activation markers include CD45RA (naive) vs CD45RO (memory), CCR7 and CD62L (central memory), CD28 (co‑stimulation). Regulatory T cells: CD4+ CD25high FOXP3+ with low CD127. Activation/exhaustion markers: CD69, HLA‑DR, CD25, PD‑1 and CTLA‑4. CXCR5 marks T follicular helper cells. These markers are used in flow cytometry and immunophenotyping.

CD62L (L‑selectin) is an adhesion receptor expressed on leukocytes that serves as a marker of naïve and central memory T cells and of lymphocyte homing to peripheral lymph nodes. Its cleavage indicates cellular activation. CD62L expression also helps distinguish leukocyte subsets (including certain neutrophil and monocyte populations) and is used clinically to assess immune cell trafficking and activation status in infections and inflammatory disease.

CD27 is a TNF-receptor family costimulatory molecule expressed on most naïve and central memory T cells; it promotes T cell survival, proliferation and differentiation after antigen encounter. Sustained CD27 expression marks less-differentiated, long-lived T cells, whereas loss of CD27 is associated with terminally differentiated effector or senescent T cells, helping distinguish memory/naïve versus effector phenotypes.

Pan-T cells generally refer to T lymphocytes identified by pan-T cell markers—surface proteins common to most T cells (for example CD2, CD3, CD5, CD7). Antibodies against these markers are used in lab tests (flow cytometry, immunohistochemistry) to detect, quantify and classify T-cell populations. The term also appears in clinical contexts like pan-T-cell lymphomas, meaning malignancies of the T-cell lineage.

The CD4 marker is a surface glycoprotein on helper T cells (and some macrophages and dendritic cells) that binds MHC class II on antigen-presenting cells. Acting as a co‑receptor, it stabilizes T‑cell receptor–MHC interactions, enhances signaling for T‑cell activation and cytokine release, and coordinates adaptive immune responses. CD4 also serves as the primary receptor exploited by HIV for cell entry.
