CD 10 / CALLA (Prognostic ALL Marker)

CD10-positive acute lymphoblastic leukemia (common B‑ALL) generally carries a favorable prognosis, particularly in children, because it is chemosensitive and achieves high initial remission rates. Long-term survival is good with modern therapy, but outcomes depend on age, white‑cell count, cytogenetic abnormalities (such as the Philadelphia chromosome) and minimal residual disease response. Adults often have more variable outcomes.

CD10 expression can have prognostic value in specific cancers—e.g., stromal CD10 often associates with worse outcomes in breast and some gastrointestinal tumors, and CD10 status helps classify certain leukemias. However, its significance varies by cancer type and context; CD10 is not a universal prognostic marker and must be interpreted alongside other clinical, histological and molecular findings.

CD10 is a cell-surface neutral endopeptidase expressed on normal germinal-center B cells, certain hematopoietic precursors and epithelial cells. Clinically it serves as a diagnostic marker for germinal-center B‑cell neoplasms (e.g., follicular lymphoma, some diffuse large B‑cell lymphomas), CALLA-positive pre‑B acute lymphoblastic leukemia, and selected carcinomas such as renal or endometrial stromal tumors.

The CALLA marker (Common Acute Lymphoblastic Leukemia Antigen), also called CD10, is a cell-surface protein on immature B lymphocytes and some other cells. Its detection by immunophenotyping helps diagnose and classify acute lymphoblastic leukemia and certain lymphomas, informs prognosis, and aids treatment planning by identifying leukemia subtype from blood or bone marrow samples.

One strong good prognostic factor in acute lymphoblastic leukaemia is a rapid early response to therapy with undetectable minimal residual disease (MRD) after induction. MRD negativity reflects effective clearance of leukemic cells and predicts lower relapse risk and better long‑term survival. Other favorable factors include age 1–10 years, low presenting white cell count, hyperdiploidy and t(12;21) (ETV6‑RUNX1).

CD10 is a cell-surface enzyme (neutral endopeptidase) expressed on early B‑cell precursors and commonly on B‑cell acute lymphoblastic leukemia (ALL). Its detection by flow cytometry confirms B‑cell lineage (“common” B‑ALL), helps classify immunophenotype, guides prognosis and treatment decisions, and aids minimal residual disease monitoring. CD10 expression is often seen in childhood ALL and can correlate with better treatment response.
