Bacterial Latex Agglutination

Latex agglutination tests detect bacterial surface antigens to rapidly identify pathogens. Common targets include Streptococcus pneumoniae, Neisseria meningitidis, Haemophilus influenzae type b, group A and B Streptococcus, and Staphylococcus aureus (via coagulase/protein A). The test detects capsular polysaccharides or specific proteins in CSF, blood, or colony material for quick presumptive diagnosis pending culture.

A bacterial agglutination test is a laboratory method that detects specific antibodies or bacterial antigens by mixing patient serum (or antigen) with known reagents; if matching antibodies are present, visible clumping (agglutination) occurs. It’s a rapid, simple tool for identifying bacterial species or immune responses, useful in diagnosis and serotyping, but requires controls and can yield false positives or negatives.

Latex agglutination uses microscopic latex beads coated with antibodies or antigens; when mixed with a patient sample containing the corresponding antigen or antibody, the beads become cross-linked and form visible clumps. Agglutination indicates a positive reaction and presence of the target molecule. The test is rapid, simple, and commonly used for pathogen identification or detection of specific serum antibodies, with appropriate controls for accuracy.

The agglutination test for meningitis is a rapid laboratory method that detects bacterial antigens in cerebrospinal fluid, serum or urine. Latex or antibody-coated particles clump when specific meningitis-causing antigens (e.g., Neisseria meningitidis, Streptococcus pneumoniae, Haemophilus influenzae type b) are present, giving a visible reaction. It aids early diagnosis but complements, not replaces, culture, Gram stain and PCR due to variable sensitivity and specificity.

Latex agglutination tests detect microbial antigens or host antibodies to diagnose infections and immune markers. Common uses: cryptococcal meningitis (Cryptococcus neoformans), pneumococcal, meningococcal and Haemophilus influenzae type b meningitis; Group A/B streptococcal and Salmonella infections; hepatitis B surface antigen; and noninfectious markers such as rheumatoid factor and C‑reactive protein, commonly used for rapid laboratory diagnosis.

A positive agglutination test shows visible clumping of particles (cells or coated beads) when a specific antigen and antibody interact. It indicates the presence of the target antigen or antibody — for example, in blood typing, infection diagnosis, or antibody screens. Results usually need clinical correlation and confirmatory testing because cross-reactions or technical errors can produce false positives.
