Anti ds-DNA Antibody - IFA

A positive anti-dsDNA test means the immune system is making antibodies against double-stranded DNA, a finding highly suggestive of systemic lupus erythematosus (SLE). Levels often track disease activity, particularly risk of kidney involvement (lupus nephritis). It isn’t diagnostic alone—results must be interpreted alongside symptoms, clinical exam and other tests (ANA, complement, urine/kidney assessments) for diagnosis and management.

dsDNA antibody by IFA is an indirect immunofluorescence test that detects autoantibodies against double-stranded DNA. Using substrates such as Crithidia luciliae, it is relatively specific for systemic lupus erythematosus and helps diagnose and monitor disease activity—higher titres often reflect flares. Sensitivity is variable, so results should be interpreted alongside clinical findings and other laboratory markers.

A normal (negative) anti-dsDNA result is assay-dependent but commonly falls below the laboratory’s cutoff—typically <25–30 IU/mL for many ELISA tests; some labs use <10 IU/mL. Crithidia luciliae immunofluorescence is reported qualitatively (negative/positive). Values above the lab reference range are considered elevated. Always interpret against the specific lab’s reference interval and clinical context.

Anti-dsDNA antibodies are autoantibodies that target double-stranded DNA. They are uncommon in other conditions but can rarely appear with certain drug‑induced autoimmune reactions and, infrequently, in other autoimmune disorders or infections. Anti‑dsDNA is used to support SLE diagnosis and to monitor disease activity, particularly renal involvement.

Early signs of lupus in females include persistent fatigue and unexplained low-grade fever; joint pain, stiffness or swelling (often in small joints); photosensitivity and a red “butterfly” rash across the cheeks and nose; hair loss and painless mouth or nose ulcers; Raynaud’s phenomenon (cold, white or blue fingers/toes); and chest pain with deep breaths from pleurisy. Symptoms often come and go.
